Summary The phytoplankton of four ponds in the lower Piedmont and upper Coastal Plain of North Carolina was studied to find a possible correlation between bound phosphorus plankton numbers. No correlation was apparent therefore this paper reports a detailed study in relation to other habitat factors.The ponds are all shallow (2 m.). The Piedmont ponds are high in colloidal clay while the Coastal Plain ponds are much lower. The pH ranged around 7.0 except for one Coastal Plain pond which remained close to pH 6.4. Water temperature ranged from 30° C in June to 23° C near the end of August.Phytoplankton numbers were determined by the standard Sedgwick-Rafter method. Populations were low except for brief pulses. One Coastal Plain pond was typical of a fertilized pond and had several tremendous blooms of blue-green algae. The others supported a seemingly typical Caledonian flora, rich in species but low in number of individual organisms.There was no clear correlation between phytoplankton populations and any habitat factor except pH. Utilizing horseradish peroxidase as a tracer protein, it is shown that trichogen and tormogen cells have a secretory function. Protein tracer from the haemolymph enters these cells by endocytosis and is transported to the sensillum liquor cavity by transport vacuoles and multivesicular bodies. We have used N-hydroxysuccinimido-d-biotin as a reagent for labeling proteins exposed at the surface of cultured bovine adrenal chromaffin cells during Ba2+-stimulated secretion. A specific secretory granule membrane constituent, dopamine-beta-hydroxylase (DBH), has been investigated using immunoprecipitation followed by electrophoresis. Within 30 min of stimulation, exposed DBH had been cleared from the cell surface. Nevertheless, quantitation of labeled DBH using [125I] streptavidin suggested that it remained undegraded over a period of 24 h, a time during which secretory granule stores of catecholamines were being replenished. Subcellular fractionation of the cultured cells suggested that, after 3 or 4 h, the biotinylated DBH, which was still membrane-bound, was located in particulate material that also contained cytochrome b561, another major secretory granule membrane component. 相似文献

Summary Dark grown coleoptile segments were floated on solutions of IAA alone and of IAA and the secretion inhibitors cytochalasin and monensin. The secretion inhibitors prevented normal elongation of the tissue segments, the monensin inhibition being virtually complete while cytochalasin gave a 40% reduction over the first six hours with little further further elongation in the following 18 hours. Vesicle production was assessed in outer epidermal cells after 6 hours of IAA-stimulated elongation using the vesicle accumulation method following a cytochalasin-block of vesicle transport. The results were compared with the area of plasma membrane required to enable cell elongation to proceed at the observed rate. The area of vesicle membrane delivered to the cell surface exceeded this requirement to such an extent that at least 65% of the delivered membrane must be recycled back into the cytoplasm. Expressed in terms of the whole cell, the plasma membrane turnover rate was found to be once every 200 minutes. It is concluded that limitation of elongation by secretion inhibitors is more likely to reflect a requirement for the vesicle contents than the vesicle membrane.
